Excited to share my thesis work!
Using super-resolution microscopy and a new HaloTag-Nup107 mouse line, we find that 1) NPC biogenesis is upregulated during neuronal maturation and that 2) torsinA is essential for normal localization and assembly kinetics of neuronal NPCs. (1/N)
TorsinA is essential for the timing and localization of neuronal nuclear pore complex biogenesis biorxiv.org/cgi/content/shor… #biorxiv_cellbio

So happy and relieved that this is now out in nature.com/articles/s41556-0…! Thanks to all authors esp Tri, @ShahmoraLab for help w revisions, and mentors @BarmadaLab, @pappassa, and @BillDauerNeuro for continued support. Also thanks to the reviewers who had thoughtful suggestions.
Very proud of Ava on her successful summer project and poster presentation! 😄
Congrats to Ava @AvaConyer on a fantastic MSRP poster presentation on the polycomb work she did together with Sumin @suminkim28 !
Very neat approach and beautiful movies!
Excited to share our new preprint with @BuvanSR and @volodya_gelfand! 🎉 Using a sparse vimentin SunTag labeling strategy and 3D FIB-SEM reconstructions, we do a deep dive on vimentin IF dynamics and nanoscale organization in cells.

You may know ZNF143 as a looping factor. We show together with the @hansen_lab that this is not the case! If you want to know why and what it does instead do read our thread below:

(1/14) Excited to share our new work on ZNF143/ZFP143 by @DomenicNarducci
ZNF143 has been widely reported to be a general loop factor
Instead we and @deWitLab in a companion preprint show that ZNF143 is a TF with no loop function:

(1/n) Excited to share our new review by @JinHarveyYang @NatRevMCB
We discuss:
1) spatiotemporal mechanisms of E-P interactions
2) mechanisms of enhancer selectivity
3) pros and cons of 3C and imaging based methods

Come and join us!
We are looking to recruit a Research Assistant/Technical Associate I to join our team Summer 2024.
This could be an ideal position for someone looking for 2 years of research experience before starting graduate school.

RT if you don't mind!! ~~ Does anyone know of a genome-wide genetic screen, or proteomics dissection, targeting for molecules required for nuclear envelope breakdown in proliferating cells of animals?
Any thoughts appreciated!!

I’m excited to share that my PhD paper is now published in @NatureComms! I’m grateful for the support of all collaborators, in particular my advisor @shiner80, Alessia Loffreda, and Emanuele Colombo. See this thread for more details, and read the paper at nature.com/articles/s41467-0…
...And the manuscript is now out in @NatureComms: nature.com/articles/s41467-0…. Here a long-due but (hopefully) short thread:
Super excited to start as a postdoc in @hansen_lab today! Looking forward to working with this fantastic group of scientists, and the view is pretty great too ☺️
Excited to share that I defended my thesis today and became the first Dr. in my family! 😊 Many many thanks to my mentors @BillDauerNeuro and @BarmadaLab!
Had a fantastic time at #celldyn2023, a big thanks to the organizers and @Co_Biologists! Feeling energized after meeting so many awesome scientists, and loved exploring the beautiful Lisboa and surroundings ☺️

Replying to @ChristinaAdele6
@ChristinaAdele6 @lab_ogden kicked off the final session of #celldyn2023 dissecting the mechanism of SHH-dependent cytoneme formation. Next up, @suminkim28 showed the role for TorsinA in neuronal nuclear pore biogenesis with disease implications.

(1/n) Excited to share preprint by @ViraatGoel @mileshuseyin
We develop Region-Capture Micro-C (RCMC)
By focusing on specific regions, we generate the deepest 3D genome maps thus far
We see coalescence of enhancer and promoters into “microcompartments”
biorxiv.org/content/10.1101/…
thanks Bill!
Congrats to @suminkim28 who led this cool work revealing the dynamics of the neuronal nuclear pore and identified a novel aspect of torsinA biology. Hat tip also to @pappassa who played essential role biorxiv.org/content/10.1101/… @UTSWBrain @UTSWNeurosci
As for the functional consequences: delayed NPC assembly may interfere with neurodevelopmental processes that depend on transport, and abnormal NPC localization may disrupt gene expression since nups have roles in genome organization. (12/N)
We hope this work provides a framework for future NPC studies in neurons & the tools will be adapted in future physiological and disease studies. Thanks to everyone who made this work possible, especially @VeatchLab and Sebastien Phan (NCMIR, UCSD) for collaborations! (13/13)
