Out today from the Anderson lab, Oblique line scan microscopy achieves nanoscale spatial and sub-millisecond temporal resolution across a large FOV, enabling robust single-molecule biophysical measurements and tracking in both cells and solution. nature.com/articles/s41592-0…
We often speak about chromatin as being accessible or inaccessible, but what does it mean? We wrote a short review on this, 🔬 focused:

A big thank you to Tom Fillot for his efforts on this and to @hansen_lab @marcelonollmann for their help as editors.

A dream come true: in 2004 I started thinking about counting microstates to predict gene expression with @JKondev Rob Phillips @HernanGGarcia. 20 years later we can measure them: nature.com/articles/s41586-0… Thank you @WJGreenleaf @bgrdoughty @MichaelaThinks Julia and all co-authors!

7 years ago, I met a junior fellow named @JD_Buenrostro who blew me away with a vision of futuristic genomic technologies
Today, we (@ajaylabade31, @carolinecomenho) are excited to share our first steps into that future: Expansion in situ genome sequencing

Fantastic, fantastic work! Congratulations @MustafaMir16 and team!
1/n) Excited to share our new preprint: Using high-res live embryo imaging we show that transcription factor hubs are localized to new genes when specific DNA binding is perturbed. Hub interaction kinetics at these new sites determine factor occupancy: biorxiv.org/content/10.1101/…
ALERT: We are looking for post-doc candidates for an AIRC funded project where we propose to track the p53 oncosuppressor as it travels across and between nuclear comparments. Plese RT!

Our work using single-molecule chromatin configurations to link TF binding to gene expression is now out on bioRxiv! Amazing effort from @bgrdoughty, @MichaelaThinks, and @juliaschaepe, and an immensely rewarding collaboration with the @BintuLab: biorxiv.org/content/10.1101/… [1/n]
What a great progress for fast, reproducible and large fov single molecule imaging and tracking. Fantastic preprint by @xyzDrch @Dan_J_Anderson @XDarzacq @Eric_Betzig and all the team @EikonTX. Now I want an OLS microscope!
Single-molecule tracking with Oblique Line Scan Illumination:
FOV: 250x190 µm🔬🦣
Frame rate: up to 1250 Hz✈️
Resolution: ~15 nm🎯
Cool preprint by @xyzDrch @Dan_J_Anderson et al. @EikonTX @TjianDarzacq @HHMIJanelia

Our first paper is out in @CellCellPress! Led by @born2raisecell, we present a programmable reaction-diffusion system that generates synthetic protein waves, oscillations, and patterns for spatiotemporal circuit design and FM data-encoding in human cells. authors.elsevier.com/a/1iN8Z…
A fantastic work from my dear science buddy @SamuZambrano, and the talented PhD student @KizilirmakCise. How do 'quasi-identical' clonal cells display heterogeneous protein dynamics? Here the focus is on NF-kB, yet methods and implications have broad applicability.
Happy to see online the work led by the talented @KizilirmakCise on how the heterogeneous NF-κB signalling dynamics emerges in a homogeneous cell population. Link below and short thread ahead. sciencedirect.com/science/ar….

Cool that with all the hectic dynamics of p53 PMl Bodies and condensed Chromatin are completely immobile.

Thrilled to share the work from my PhD and @BernardiLab now out on @NAR_Open. Here we dissected the multiple and parallel transcriptional functions of #PML defining it as an important regulator of the epigenome in triple-negative breast cancer.

...And the manuscript is now out in @NatureComms: nature.com/articles/s41467-0…. Here a long-due but (hopefully) short thread:
Proud to share our latest preprint: we explore how chromatin organization and transcription factor search are related using a freshly built 🔬, to combine single molecule tracking of nuclear factors with structured illumination imaging of DNA. biorxiv.org/content/10.1101/…
7/ Our data highlight that chromatin can influence the nuclear factor search mechanism and that modulating interactions mediated by the factors IDRs can divert such search. We hope to extend the SMT/SIM approach to probe NF dynamics and activity at different nuclear compartments:
All kudos @mazzocca_matteo and Alessia Loffreda, who drove the project, to Zeno Lavagnino and Emanuele Colombo who built and validated the 🔬, to all the other members of the lab that contributed in so many ways, and to our collaborators @LabLegube and Edouard Bertrand lab.
