Our paper is out in @Nature! Extrachromosomal DNA molecules huddle together in cancer cells to activate oncogene expression (which we call ecDNA hubs). A great collaboration with @KatieEYost, @HowardYChang, @549nm, Zhe Liu, and others. (1/10) nature.com/articles/s41586-0…

Our study "Individualized VDJ recombination predisposes the available Ig sequence space" is now published in Genome Research 🥳

Congrats @rlyhighvariance et al.!
1/11 Did you ever wonder whether individuals have the same probability of generating a given antibody sequence? Turns out, they do not – not even twins!
We showed that humans – including monozygotic twins – have unique VDJ recombination rules. biorxiv.org/content/10.1101/…

1/Excited to share my first author publication where we create split-engineered base editors (seBEs). Here, we place base editors under the control of a small-molecule to regulate the timing of genome editing and decrease off-target effects. To achieve it: nature.com/articles/s41589-0…

Accepted manuscript strongly suggesting that apparent SARS-CoV-2 reads that appear to be integrated in the human genome, are in fact an artefact of library preparation.

There is an important message here for today’s students. Techniques don’t matter. Questions do. Listing a set of techniques is silly because they will all be out of date in 10 years. But questions drive science forward.

If you’re having a hard time with glasses fogging or keeping your mask up over your nose, a simple bandaid does wonders. Learned it in the OR.

This new mink COVID-19 mutation story, which is making the rounds on Twitter is highly problematic. There are elements of truth in it but the reporting is completely irresponsible.

I'm still in shock. Alekos was a real Scientist, with a passion for RNA editing and a curiosity for everything, bio and beyond. He was one of the most generous humans I know - with his time, his possessions, his mind. I wish I had said goodbye. Até sempre Alekos.

We identified a pattern of #mutation clustering in cancer #genomes, distinct from the known 'mutation showers'⛈️
We think one cause of such 'mutation fog'🌫️is DNA repair + APOBEC3A activity

ALT We classified the diverse patterns of clustered mutagenesis in tumor genomes, which identified a new A3 pattern: nonrecurrent, diffuse hypermutation (omikli). This mechanism occurs independently of the known focal hypermutation (kataegis), and is associated with activity of the DNA mismatch-repair pathway, which can provide the single-stranded DNA substrate needed by A3, and contributes to a substantial proportion of A3 mutations genome wide.
