For >10 years, neutrophils have challenged a central assumption of inflammasome biology by secreting IL-1 without robust pyroptosis. Our new study shows that the local inflammatory milieu regulates neutrophil susceptibility to CASP1-dependent pyroptosis.

Patients can look well-controlled and still have epithelial death programs running. We found it in nascent IBD lesions on advanced therapy and it flagged who relapsed over 2–3 years. @WEHI_research @NHMRC #ScienceResearch
Congratulations to our co-first authors See Jie Yow, Bing-Chen Wu, and Pung Hai Shin, and sincere thanks to all co-authors and collaborators for their contributions to this work!

How lytic cell death drives autoantigen release and fuels inflammation? With @lynndustinlab we ask this using a common autoantigen, TRIM21. The autoantigen TRIM21 assembles proinflammatory immune complexes after lytic cell death | Science Immunology science.org/eprint/VEVN4EQK8…

TRIFosomes found😀. Very proud of our team’s work to identify endogenous TRIFosome. Surprisingly whilst TRIFosomes activate NFKB, IFN signaling is from monomeric or dimeric TRIF. doi.org/10.1126/sciadv.aeb95…

Inflammatory signalling must eventually be turned off (e.g. by feedback inhibition) to prevent chronic inflammation. We didn't know how #inflammasomes turn off their signalling in vivo ... until now! (🧵: 1/3)

The loss of TBK1, or both TBK1 and the related kinase IKKε, results in uncontrolled cell death–driven inflammation. We show that TBK1/IKKε prevent premature cell death by limiting the activity of multiple death pathways in myeloid cells.@FabiAlexFischer science.org/doi/10.1126/scia…
