#daraxonrasib clinical outcomes by allele in #RASolute302 from FDA NDA review. These data perpetuate unresolved questions: - is RASi as effective for KRAS WT; K12R? See Table 24; Fig 11. - is 300 mg necessary for maximal clinical benefit? no data provided comparing 200 vs 300.

The incidence of early-onset colorectal cancer has doubled in the last 25 years. This is a fantastic @NEJM review article on #EOCRC that I stumbled across today. It's 5 years old, but @FASinicropeMD summarizes the topic wonderfully. It covers risk factors, preventative factors, and the role of the microbiome. The graphics are excellent. Smoking and obesity may be obvious contributors, but there's also an alarming association with the Western diet (red meat, ultra-processed foods, etc). The trends in this review stop at 2016. A scary revelation is that incidence and annual percent change have kept going up since then: 2016: incidence 8.1 per 100k, APC 3.4% 2023: 11.5 per 100k, APC 9.9% Article: nejm.org/doi/full/10.1056/NE… SEER explorer: seer.cancer.gov/statistics-n… Systematic reviews: pubmed.ncbi.nlm.nih.gov/4219… pubmed.ncbi.nlm.nih.gov/3722… Pooled analysis of 3 large European cohorts: pubmed.ncbi.nlm.nih.gov/4142…
