8/ Together, our study shows that RA orchestrates immune memory across tissues, guiding tissue-residency and long-term surveillance. Harnessing RA’s potential could help us design therapies to boost immune protection and manage autoimmunity. 🚀
5/ We also found that RA + TGF-β work together to establish gut TRM identity. This is something we recapitulated in a dish in collaboration with @colbyzaph, whereby T cultured cells with these signals adopted TRM-like traits. 🔬
4/ The answer: T-bet controls sensitivity to retinoic acid (RA), a vitamin A metabolite essential for licensing TRM development in the gut (recently shown by @BSheridanLab). Meanwhile, liver TRM also rely on RA—instead of TGF-β—to guide their formation.
3/ In skin and liver, we previously found that T-bet enables TRM to respond to IL-15. But gut TRM thrive without IL-15 (as @JMSchenkel & @Masopust_Vezys reported). So what makes gut TRM tick?
2/ TRM protect tissues like the gut, skin, and liver, but how do they establish residency in such diverse environments? 🤔That’s the key question we tackled. #TeamTRM
