Paper alert! 📢 PHYCUT: multiplexed Cas9 editing for microalgae. pubs.acs.org/doi/10.1021/acs… Amazing work from #EmilyStuckless with assist from #Gloor_lab. Multiplexed knockouts of fucosyltransferase genes for humanization of N-linked glycosylation pathway.
🔊Paper alert! Incredible work from #EmilyStuckless and fellow Edgell lab members on PHYCUT multiplex Cas9 editing system for Phaeodactylum tricornutum. biorxiv.org/content/10.1101/…
PHYCUT used for glycoengineering by knocking out fucosyltransferases as first step in humanizing N-linked glycosylation pathway!

Paper 🚨📢! Adenine methylation in PAM sequence regulates Staph aureus Cas9 activity in bacteria. biorxiv.org/content/10.1101/…. Superstar collab between #DaltonHam @nortnlentivirus
crisprHAL ML model found poor activity at PAM sites with GATC sequences....seem familiar? GATC is DNA adenine methyltransferase site in bacteria. Large scale in vivo assays, Nanopore sequencing, and in vitro assays all show that G*ATC methylation impacts SaCas9 activity.
All the details in the paper! Another stellar collaboration with #GloorLab. Supported by @CIHR_IRSC @westernuBiochem @SchulichMedDent

4/n Guess what? Size does matter - everything fits within a single AAV vector (TevSaCas9 + repair guide). NO need for co-delivery of exogenous repair factors, accessory repair proteins/templates, or small/molecules/proteins to modify repair pathways.
