👉 The oncogenic #EGFR–SHC1 #fusion drives hashtag#lungcancer through dual activation of the N-terminal EGFR kinase domain and C-terminal SHC1 phosphorylation sites. This biology requires combined inhibition of both EGFR and SRC-family kinases to achieve effective clinical control.
👉 Most importantly, this represents a novel fusion activation mechanism: SHC1 acts not merely as a facilitating #fusion partner, but as a true #collaborating partner in oncogenesis: a previously unrecognized mechanism with direct therapeutic implications.
👉 Could similar cooperative signaling by fusion partners help explain de novo or early resistance in other fusion-driven cancers? 😜

High quality study from @BenjaminBesseMD showing only 26% of pts need retreatment after getting 2 yrs pembro for NSCLC.
However, outcome for IO rechallenge was mixed, w/ many pts clearly not responding and dying. Much like CM-153 & w/ prior pooled report of Abreu-Rodriguez showing only 19-27% ORR with pembro re-challenge.
So still an unresolved question: whether 2-yr discontinuation is optimal for all patients?

ALT Kaplan-Meier graphs show overall survival and time to next treatment. Graph A shows overall survival probability decreasing over 48 months, with numbers at risk decreasing from 1480 to 46. Graph B shows the same trend for time to next treatment/death, decreasing from 1480 to 62.

Based on the current CiteScoreTracker 2025, the journal is trending around ~3.6—a very encouraging signal and a great achievement for a young, focused journal in liquid biopsy and precision oncology.

A topic close to my heart for years... family history in #NSCLC is often underestimated and diffucult to ascertain given the role of other stronger risk factors (smoking/environmental)
In FAHIC-Lung, our team at @CampusBioMedico and @PoliclinCampus developed a structured, weighted family history score identifying patients with enriched familial cancer risk, to support targeted referral to genetic counseling & germline testing.
