🔴 What if different infections are driving the same cycle of immune dysfunction?
A new study has revealed persistent activation and exhaustion-like dysfunction of virus-specific CD8+ T cells in long COVID, alongside altered immune responses to Epstein–Barr virus (EBV).
These findings raise some fascinating questions.
Why do infections caused by completely different viruses sometimes lead to remarkably similar chronic illnesses, such as ME/CFS and long COVID?
Could persistent viral antigens keep the immune system activated for months or even years, gradually impairing T cell function and making it increasingly difficult to control infections such as EBV?
And could this prolonged immune dysfunction also contribute to the development or maintenance of autoimmunity?
These are questions we have been exploring in our research since 2021.
In 2023, we extended our immunopathological model to long COVID, proposing that persistent antigenic stimulation, inadequate viral control, altered T cell differentiation, and immune exhaustion could contribute to a self-perpetuating cycle of immune dysfunction.
The new findings are consistent with several important aspects of this model, although they do not establish that ongoing viral replication is responsible for the observed abnormalities.
But perhaps the most important question is what all of this means for treatment.
If a persistent viral infection is helping sustain chronic immune activation, T cell dysfunction, and potentially autoimmunity, shouldn’t controlling that infection with appropriate antivirals be one of our therapeutic priorities?
After all, restoring immune function may be difficult if the infectious stimulus contributing to its dysfunction remains present.
This does not mean that every patient with ME/CFS or long COVID needs antivirals. It means that identifying patients with demonstrable persistent viral replication or clinically relevant viral reactivation should become an important research and therapeutic priority.
And importantly, persistent viral antigens and active viral replication are not necessarily the same thing. Understanding that distinction will be essential for determining which patients might actually benefit from antiviral treatment.
Could targeting persistent infections help break the cycle of immune exhaustion and chronic inflammation? And are we getting closer to a more targeted antiviral approach for these diseases?
I have explored these questions in a new, more detailed article, connecting the latest findings with our previous research and discussing their potential implications for ME/CFS, long COVID, autoimmunity, and future antiviral strategies.
I genuinely enjoy exploring these connections and making complex immunology accessible, especially when the research could eventually help us understand and treat diseases that have been neglected for far too long.

People with food allergies were shown to have elevated gut histamine, normal or slightly lowered DAO, but significantly lowered HNMT.
DAO is the enzyme that breaks down extracellular histamine.
HNMT breaks down histamine intracellular.
This raises questions about whether people with food allergies have issues with histamine clearance.
But also, why would someone with an allergy have down-regulated HNMT?
Perhaps issues with SAH, or methyl donors. Perhaps it relates to genetics or even leaky gut/inflammation.

These are my babies! I can tell you that we are still organic and a lot of our digestive issues remain improved. My kids are now doing great in high school
This doctor just revealed the shocking truth about glyphosate and autism.
Bookmark this.
“The correlation coefficient is 0.99.”
“It’s almost an absolute correlation.”
Then, he revealed how one mom cut her kids’ glyphosate exposure by “94.2%.”
She has triplets.
Two have autism and one has seizures.
All three had “high values of glyphosate.”
Why?
“She was giving corn tortillas for breakfast.”
“Corn tortillas for lunch.”
“Corn tortillas at dinnertime.”
“Corn tortillas for bedtime snack.”
Dr. William Shaw: “The kids were getting massive amounts of glyphosate from the corn.”
“When I told the mother … she immediately stopped the food.”
“She put them on an organic diet.”
“In about two months, there was a 94.2% reduction in glyphosate.”
“The most common question was what kind of therapy can I give my child to get rid of this if they have glyphosate?”
“The only therapy you need to do is to use only organic food.”
“Don’t use any standard food.”

French President Emmanuel Macron left France for a three day Gulf tour. 🇫🇷
He is governing a country where the prime minister is calling the absence of deaths “a miracle,” where a 17 year old lost an eye and a student lost a hand, and where his own interior minister says authorities “lost control.”
French public just watched 2,400 of their fellow citizens get arrested, watched the base of the Marianne statue get torn apart and the president’s response was to get on a plane. Whatever Macron secures in the Gulf this week, he will come home to a country that watched him leave when it needed him most.

This week I went to the pharmacy to pick up more LDN and ended up chatting with the pharmacist about MCAS, EDS, and POTS.
She told me that all of her MCAS patients would likely need to take antihistamines daily for the rest of their lives. Lol? She also insisted that MCAS is purely genetic and spoke about it as though lifelong pharmaceutical management was essentially inevitable.
I politely mentioned that I personally know several people with MCAS who have been able to come off antihistamines, manage their symptoms through other approaches, and experience significant improvements in their health.
Her response? “Well, they probably don’t have MCAS then.” 😂 Apparently, improving to the point of no longer needing daily medication calls your original diagnosis into question?
She proceeded to speak to me in an increasingly condescending manner, dismissing anything that challenged her understanding of the condition, until I eventually just walked out. Very strange interaction.
It’s so concerning that “medical professionals” present one approach to managing a complex condition as though it is the only possible trajectory. This is particularly concerning with conditions like MCAS.
I don’t know why I would expect anything different, but I found myself a bit shocked by this. Another beautiful reminder of how important it is to advocate for yourself and develop your own understanding of your health. You must be discerning about whose understanding of health you allow to shape your own. A medical professional’s interpretation is not an infallible truth, and a diagnosis should never become a story about the limitations of what your body is capable of.

Why Ketotifen, Vilon, And Quercefit Fix 90% Of Chronic MCAS And Histamine Flares In Some People While Doing Nothing For Others
People spend thousands on functional doctors, immunology panels, and mast cell stabilizers only to watch this exact three-compound stack stall out or worsen their flares.
Stabilizing mast cells fails when practitioners hand out compounds like a checklist while ignoring the mineral, methylation, and nuclear cofactors required to execute the biochemistry and clear trapped histamine.
1. Ketotifen (Locking Mast Cell Blast Doors And The Magnesium Trap)
Ketotifen clamps mast cell membranes shut, but it cannot hold the line if your intracellular energy and liver clearance pathways are starved.
Plugging The Calcium Ignition Switch (H1 Inverse Agonism and cAMP-PDE Inhibition): Think of a mast cell like a grenade that pulls its pin when calcium floods in; Ketotifen locks H1 receptors and preserves cyclic AMP (via phosphodiesterase inhibition) to keep histamine sealed, a process that stalls without abundant intracellular Magnesium-ATP.
The Rebound Histamine Pool (UGT1A4 Glucuronidation and Anticholinergic Stasis): Taking Ketotifen before mapping hepatic Phase II glucuronidation (UGT1A4) and vagal tone slows gut motility while uncleared histamine pools outside receptors, triggering lethargy, weight gain and rebound flares.
2. Indena Quercefit (Disarming The Allergy Trigger And The COMT Jam)
Standard quercetin washes out in stool, whereas Indena Quercefit wraps quercetin in a sunflower phospholipid bubble for 20-fold absorption to halt histamine synthesis.
Cutting The Ignition Wire (Fc-Epsilon-RI Syk Kinase and HDC Inhibition): Quercefit cuts the wire connecting allergy antibodies to degranulation (inhibiting Syk kinase and PLC-gamma downstream of Fc-epsilon-RI) and blocks Histidine Decarboxylase (HDC), but oxidized quercetin irritates cells unless intracellular Vitamin C and reduced glutathione recycle it.
Clogging The Methylation Drain (COMT Saturation and Catechol Pooling): Because quercetin exits through your liver's methylation drain (COMT), dosing Quercefit while depleted in Magnesium and SAMe (from riboflavin, folate, or choline deficits) jams COMT, causing adrenaline and estrogens to back up into wired insomnia.
3. Vilon Bioregulator (Rebuilding Thymus Brakes And The Zinc-Retinol Lock)
While Ketotifen and Quercefit muzzle mast cells temporarily, Vilon enters your shrunken thymus gland to rebuild the immune peacekeepers that turn MCAS off upstream.
Unzipping Dormant Immune DNA (PEPT1 Transport and Histone H1 Remodeling): Vilon (a Lys-Glu dipeptide) slips through intestinal PEPT1 doors to unspool knotted thymus DNA (via histone H1 binding), reactivating IL-2 and FOXP3 transcription to produce peacekeeper Regulatory T-cells (Tregs) that dock onto mast cells (via OX40-OX40L) and command them to stand down.
Empty Key Blanks In The Thymus (Zinc-Thymulin and RAR-RXR Silence): Vilon fails if Thymulin lacks its intracellular Zinc spark plug (to fold the active Zinc-Thymulin complex), or if new Tregs lack cellular Vitamin A (retinoic acid) and Vitamin D3 to unlock nuclear RAR, RXR, and VDR receptors.
4. The Enzymatic Drain (Why Stabilizers Fail Without DAO And HNMT Cofactors)
Locking mast cells shut still leaves you flaring if the enzymatic drains clearing existing histamine lack their vitamin and mineral spark plugs.
Unplugging The Gut Drain (Bioavailable Copper and P5P for DAO): Gut Diamine Oxidase (DAO) acts like a stalled blender without bioavailable Copper (via hepatic ceruloplasmin) and active Vitamin B6 (P5P), yet dosing them out of sequence over a congested liver triggers neuropathy and oxidative flares.
Unplugging The Brain Drain (SAMe and Molybdenum for HNMT and SUOX): Brain histamine clearance requires HNMT transferring methyl groups from SAMe, meaning unmapped methylation blocks or molybdenum-depleted Sulfite Oxidase (SUOX) pathways trap central histamine and drive migraines and a high pulse.
5. Blind Self-Experimentation Liabilities
Piecing an MCAS protocol together through trial and error wastes months and degrades your baseline in an unmapped operational environment.
Molecular Form And Sequence Failure: Generic quercetin fails compared to Quercefit Phytosome, dietary protein blocks oral Vilon PEPT1 uptake, and taking Ketotifen before opening clearance pathways causes anticholinergic lethargy and rebound sensitivity.
Secondary Cofactor Depletion: Pushing mast cell and thymus repair drains intracellular Zinc, Magnesium, Riboflavin (FAD), and SAMe while stripping bioavailable Copper, shutting down endogenous DAO and Complex IV mitochondrial energy.
Unmapped Clearance Bottlenecks: Sluggish hepatic Phase II glucuronidation, sulfation (SULT1A1), and biliary stasis recirculate unbound histamine, mycotoxins, and lipopolysaccharides (LPS) through a congested internal system, making a custom lab and case audit the fastest, safest way to fix the problem once and for all.
Every internal system operates under unique constraints. What resolved MCAS, histamine flares, and allergies for someone else can easily trigger severe rebound degranulation, wired insomnia, or stall out your baseline.
DM me if you want an MCAS and histamine protocol optimized for your body specifically based on multiple variables or some other bottleneck for the exact step-by-step guide.

Update 1: (For MCAS and Long Covid People)
Got 500 ibudilast 10mg capsules and took 30mg first dose. Cognition-wise, mildly better focus, but it could be placebo so I can't say for now. No side effects, which is a good start. I will do 30mg for 1 week, then week by week increase the dose up to 120mg per day (split in 3) and write down each day what doses did what, and then settle at the dose that increases cognition without causing side effects.
For MCAS and Long COVID, Japan has a Solution called Ketas (ibudilast) since 1989. The same thing is now in Western trials for neuroinflammatory disease.
Ketas is a brain-penetrant inhibitor of PDE4 (phosphodiesterase 4, the enzyme that breaks down cAMP in immune cells) and PDE10 (phosphodiesterase 10, which breaks down cAMP and cGMP in neurons), with additional activity at PDE3 and PDE11, two more enzymes that clear the same cyclic nucleotides.
It is also an allosteric inhibitor of MIF (macrophage migration inhibitory factor, a cytokine that keeps inflammation from resolving) and an antagonist at TLR4 (toll-like receptor 4, the sensor for endotoxin and damage signals).
Japan approved it for asthma and for dizziness tied to poor cerebral blood flow. The FDA has not approved it. MediciNova runs it as MN-166, with Fast Track and orphan status in ALS (amyotrophic lateral sclerosis), and it has been taken into lingering-COVID protocols for the same glial and cytokine reasons.
MCAS:
Mast cells keep cAMP (cyclic adenosine monophosphate, the signal that tells the cell to stay quiet) low through PDE4.
Low cAMP favors degranulation, TNF (tumor necrosis factor, a core inflammatory cytokine), IL-6 (interleukin-6, the fever and fatigue cytokine), and the leukotriene arm.
Ibudilast raises cAMP by blocking that enzyme, so the cell is less willing to dump. Older pharmacology also gives it leukotriene-receptor antagonism, which is the part an H1 blocker (histamine H1 receptor blocker) never covers.
MIF is released by mast cells and then calls more inflammation in. Blocking MIF shortens that loop. TLR4 is how endotoxin and mold fragments prime the mast cell before a food arrives.
Ibudilast antagonizes TLR4 and cuts the MyD88 (the adaptor that carries the TLR4 signal inward) and NF-kB (nuclear factor kappa B, the transcription switch for inflammatory genes) traffic underneath it.
Long COVID:
The persistent form is not a missing antihistamine. It is microglia that stayed on, TLR4 still ligated by residual inflammatory signals, and mast cells firing on a lowered threshold. Ibudilast crosses into the brain, suppresses glial activation, and raises cerebral blood flow.
That is the cognitive half: processing speed and the wired-but-exhausted feeling track perfusion and glial noise more than they track dopamine.
MIF and TLR4 are the systemic half. They keep the cytokine set point high after the virus is gone. PDE4 blockade in immune cells pulls TNF, IL-1β (interleukin-1 beta, the inflammasome cytokine), and IL-6 down.
People whose long COVID looks like MCAS, POTS-adjacent flares (postural orthostatic tachycardia, the heart-rate jump on standing), brain fog, and post-exertional inflammatory crashes are the overlap this molecule was built for, even though the Japanese label never used those words.
