Just realized George Church, who I know well and is the Harvard/MIT genetics legend, CRISPR pioneer, and one of the most important scientists alive, is raising for his gene therapy company Rejuvenate Bio on Wefunder.
This is equity crowdfunding under Regulation CF. Everyday people (not just VCs or the ultra-rich) can invest as little as a few hundred dollars and own actual equity in a company spun out of his lab. One-shot AAV therapies that have already shown heart disease reversal in dogs, with years of safety data, and a path toward humans.
For most of history, this kind of science was locked behind elite institutions and big checks. Now a founder of Church’s stature and his partners are deliberately opening the round to the broader community, and it’s working (oversubscribed, ranked #1 by Kingscrowd).
Rejuvenate Bio spun out of my lab at Harvard, and I co-founded it. One injection reverses cardiac disease in dogs, with three years of safety data and zero safety issues. They’ve raised $3.2M+ and 1,250+ investors are in, and Kingscrowd ranks the round #1 in the country. It closes soon: wefunder.com/rejuvenatebio

we might be celebrating birthdays backwards soon
george church's lab just published a single gene that makes old cells act young
- they trained a model to guess a cell's age from its gene expression
- then screened a gene library for anything that made old cells score younger
- the winner was SRSF1, an RNA splicing factor
- gave it to old mice and they lived 14% longer (median), delivered to the liver
- put it in old human skin cells and senescence dropped, DNA repair switched back on
- and the cells stayed skin cells with no identity loss
so aging might be a splicing problem, and a model found the patch

Jennifer Doudna did not just discover how #CRISPR-Cas9 finds its target and invent gene editing with it. Her leadership of the @igisci has taken this technology into real world innovation and impact in disease therapies, agriculture, and climate change. Jennifer is also one of THE best science communicators in the world. I promise, if you're interested in gene editing - you'll enjoy every second.

of course that's your contention. you're a first-year frontier bio guy. you just finished reading about CRISPR, probably watched a Doudna talk too. so now you're convinced everything is just gene editing.
that's gonna last until next month when you discover base editing, and then you're gonna be talkin' about how cutting DNA was barbaric and double-strand breaks are a legacy approach.
then you're gonna read about delivery, and suddenly everything's about lipid nanoparticles and how editing was the easy part and the whole goddamn thing is bottlenecked by getting it to the right organ.
that'll last until somebody shows you AlphaFold, and then you're gonna be in here regurgitating DeepMind, talkin' about de novo enzymes and virtual cells and how wet labs are economically obsolete.
then six months from now you'll watch one assay fail four times in a real lab, and start telling everybody the bottleneck was never ideas, it's validation.
somewhere in there you'll find a guy named Keith on Telegram, and you'll start doing reconstitution math in your head at dinner.
and by next year you're gonna be standing right here explaining to me you reversed your biological age by six years because you picked the clock that said so.

WOW! 73%!!!
$ARWR just silenced a gene in the human brain with a shot under the skin. Nobody has ever done that. Not once. Not anywhere.
Here's the number nobody's reading: Day 105 is ninety days after the last dose. That's not a peak. That's a floor that held for three months with no drug on board.
Biogen gets 50-65% through a needle in the spine.
Arrowhead got 73% through a needle in the arm.
Two doses. Not three.
PSP and FTD regulatory talks already underway.
The brain had no book. Arrowhead just wrote the first page.
Today we announced interim Phase 1/2a clinical data for ARO-MAPT.
The results represent an important step in clinical validation of our TRiM platform for delivering siRNA to the central nervous system.

Thank you to @FDADrugs for making ANKTIVA accessible to all patients in non muscle invasive bladder cancer who failed BCG therapy. @ImmunityBio - The first treatment approved for this group of patients.
An important step for cancer immunotherapy 2.0.
Big news for bladder cancer patients! FDA just expanded approval of Anktiva with BCG for adults with BCG-unresponsive, high-grade papillary-only NMIBC — the first treatment approved for this group. This is a first, and that matters. Evidence from a closely related form of bladder cancer supported the approval.

$ARWR silenced a gene in the human brain with a shot under the skin. Mean 73% tau reduction, 85% in one person.
My reaction note is up.
The headline is 73%. The number that matters is 105, the study day. Last dose was Day 15, so that was measured ninety days after the drug stopped.
A primate comparison Arrowhead published in May that nobody has connected to today: spinal canal delivery varied 22x across brain regions. Under the skin, under 4x.
