Despite multiple checks and reasonably precise alignments, I think my setup isn't performing well in terms of axial resolution and light sheet sectioning. In high NA illumination, it's usually about 1.3µm, in the red channel. We expect less than 1µm!
Any hidden alignment faults or common errors I should consider to rectify this? Any raw datasets from beads in gel and 3D PSFs you can share to show how a practically good PSF should look (at high NA illumination)?
