It is debated whether poor #blood flow underlies white matter loss in #aging. Our recent study in aged mice supports this idea: Impaired drainage through rare, deep-reaching #venules causes mild #hypoperfusion that triggers inflammation and demyelination.

What makes cerebral white matter more vulnerable with age? In this mouse study, we show that large and rare venules are bottlenecks for blood flow in the white matter. Deep branches of these venules are constricted with age, leading to reduced blood flow.

Mural cell calcium imaging revealed a de-coupling of calcium activity from vessel dynamics in transitional vessels. So, mural cells are signaling but fail to translate these signals into contraction/relaxation.
Capillary injuries and regression are hallmarks of many neurological diseases, but they are exceedingly small and evade detection by most clinical imaging. Studies in animal models can help us understand how small injuries produce outsized effects through their remote actions.
Another wonderful study led by post-doc and K99/R00 fellow @stephkbeee, supported by PhD student @NielsonCara and lab manager @sosamariaj.

It's all connected... In this study, we show that tiny injuries created deep in the brain's capillary bed can contribute broadly to blood flow impairment by constricting upstream supply vessels. biorxiv.org/content/10.1101/…
Focal capillary injuries were made using precision laser ablation, and the effects of these injuries were carefully monitored across vascular zones.

We team up with @drew_lab_nvc to examine anatomical and functional changes in aged mouse brain. We found that aging induces selective regional vascular and pericyte degeneration, slowed NVC response, and lower oxygen carrying capacity.
