Keith Hamilton from the @Alushin_Lab at Rockefeller University gave a deep dive into the first step of any structural biological workflow: sample preparation. Through test cases he showed the balance of what is easily doable versus preserving biology.

🎄It’s Beginning to Look a Lot Like Christmas... Under the Microscope!🔬
Why buy Christmas trees when you can "grow" one using micropatterning ( @AlveoleLab ) and @AllenInstitute 's cells with LaminB1 "ornaments"?
Happy #FluorescenceFriday !

To challenge the limits of the system, we studied conditions with many beads in the microwells. We then looked at the fraction of weak beads moving, as a function of competition strength (defined by the number of strong beads).
Alvéole retweeted
10. We then demonstrated that when there is turnover, weak structures (from beads or micropatterns) can grow when they are in competition with strong networks. Turnover enables the weakest to exist!

9. Therefore, without turnover, the actin monomer is limiting and the available monomer pool is taken over by dense structures in a “winner-take-it-all” scenario.

8. To challenge those results in another geometry, the amazing @cguerin38 developed a protocol to generate actin networks with various densities on lipidic micropatterns at the bottom of our microwells (with the Primo @AlveoleLab).

With the PRIMO device from @AlveoleLab, we have also developed a new technique for generating micropatterns of controlled geometry, with high-friction zones juxtaposed to low-friction zones. In this case, the contraction was biased toward the region with higher friction.
