Can histone deacetylation fuel acetyl-CoA metabolism?
We teamed up with Ioana Soaita @AranyLab @mzspectrum @WellenLab to answer this question.
jbc.org/article/S0021-9258(2…
We didn't detect any release into extracellular acetate, so we checked 13C-labeling. We found that up to ~10% of the acetyl-CoA pool was derived from deacetylation, and this label was transmitted downstream into (iso)butyryl-CoA!
Our data are consistent with a model where acetate is preferentially mobilized to maintain other acetylation events (such as shown recently by Mendoza et al @berger_lab @GarciaLabMS)

Is histone acetylation really a reservoir for 2-carbon units to make acetyl-CoA? We used 13C tracing to show that ... yes, but not that much. Acetylation is dynamic but contributes only a little to cellular AcCoA demand. @WellenLab @mzspectrum @trefely authors.elsevier.com/sd/arti…
