Poster Boy :). Great discussion on our Restore update @EHA_Hematology
CONGRESS | #EHA2026 | POSTER
Ciro Rinaldi shares an update from the phase II RESTORE trial evaluating elritercept + ruxolitinib in patients with #myelofibrosis and anemia (N = 38; TD, n = 23; NTD, n = 15).
Elritercept + ruxolitinib demonstrated anemia and transfusion responses. In NTD patients, 50.0% achieved mean Hgb increases of ≥1.0 g/dL over any 12-week period. In TD patients, 43.5% achieved RBC-TI ≥12 weeks by Week 36, while 65.2% achieved ≥50% reduction in RBC transfusion. Overall, mean platelet count increases of ≥30 × 10⁹/L were observed in 39.7% of patients over any 12-week period within the first 24 weeks. Among patients with baseline spleen volume ≥450 cm³, 20.8% achieved SVR ≥25% and 8.3% achieved SVR ≥35%. Across all patients, ≥50% MF-SAF-TSS reduction was achieved in 48.1% of evaluable patients (n = 27), and 18.8% of evaluable patients (n = 32) had ≥5-point improvement in PROMIS Fatigue T score by Week 24. Grade ≥3 TEAEs occurred in 60.5% and Grade ≥3 study drug-related TEAEs occurred in 23.7%. The most frequent TEAEs included thrombocytopenia, diarrhea, fatigue, anemia, ALT increased, cough, and nausea; no Grade ≥3 diarrhea was reported, and elritercept discontinuation due to TEAEs occurred in 5.3%.

CONGRESS | #ASCO26 | POSTER
Ciro Rinaldi @tamaciro presented the design of a phase II, open-label, single-arm study evaluating bomedemstat 50 mg + momelotinib 200 mg QD for up to 24 weeks in patients with cytopenic #myelofibrosis. The study plans to enroll approximately 40 patients with primary or secondary MF and baseline cytopenias, including hemoglobin < 10 g/dL, platelet count < 100 × 10⁹/L, or transfusion-dependent anemia.
The primary endpoint is the proportion of patients achieving ≥ SVR35% at Week 24, with secondary endpoints including symptom improvement, transfusion independence, safety, OS, and PFS.
