My group made a game in a weekend for #ludumdare - check it out! It's a crafting/exploration game and I think it's super cute (I did all the art). One reviewer said "i was planning on only testing the game but I got sucked in for 30 min", go try it! ldjam.com/events/ludum-dare/…

i’m a strong believer in the power of “granny hobbies” to improve your mental health (and detox from digital everything) reading, knitting, gardening, crafting, cooking, baking, board games, sewing, birdwatching, playing cards, drinking tea …

This is actually a pretty good summary of the excellent Orwell essay “Politics and the English Language” orwellfoundation.com/the-orw…
english has a cool built-in heuristic for detecting bullshit are most of the nouns and verbs being used latin/greek or germanic in origin? if germanic, the things being discussed are probably real physical actions and things: tree, walk, cow, shit, fall, wood, buy, leg, go

Hell yes! Mayor London Breed partnered with 7-Eleven on a new initiative to bring Japanese onigiri to convenience stores in San Francisco. I’m beyond thrilled! Got in line early yesterday and snagged one 🍙 Thank you #7eleven
Readers added context they thought people might want to know
This tweet is false. There is no partnership between the mayor and 7-11 and these onigiri are not being sold. The tweet author is a “conceptual artist” who created the packaging and made up the backstory. sf.eater.com/2024/3/22/2410…

From lab to approval in 11 years 🧬 And the thing is, medical applications of CRISPR are just getting started... Just yesterday, the @doudna_lab released two juicy preprints on improved delivery methodologies for CRISPR editing machinery: 1. Lung and liver editing by lipid nanoparticle delivery of a stable CRISPR-Cas9 RNP [1] 2. Engineering self-deliverable ribonucleoproteins for genome editing in the brain [2] In the first paper, they took a thermostable variant of the Cas9 enzyme from a thermophilic bacteria (Geobacillus stearothermophilus), and used directed evolution to engineer >100x greater editing efficiency. This thermostable Cas9 could be more efficiently encapsulated in LNPs, meaning that the actual protein could be delivered in a scalable vector instead of mRNA encoding the protein. This has potential benefits for avoiding immune activation and having potentially higher editing efficiency—the translation efficiency of the mRNA can be a bottleneck. They demonstrated in vivo lung and liver targeting (@justsaysinmice). The second preprint was also very cool. What if CRISPR could just deliver itself (almost like an antibody or other biologic) instead of being packaged in the first place? To achieve this, they screened for self-delivering Cas9 proteins that were fused to cell-penetrating peptides (CPPs) that could help escort them into cells. They showed some POC data for in vivo delivery into the brain via injection into mice. It's absolutely insane to see the pace of progress for CRISPR therapeutics. From basic research result to a new approved modality in 11 years. Just a day before the approval, a pair of brand new delivery strategies released from a single lab. Genomic medicine is only just getting started... What a time to be a biologist! [1] doi.org/10.1101/2023.11.15.5… [2] doi.org/10.1101/2023.11.15.5…

A reality that most devs working at large companies take time to appreciate: Personal connections tend to make things easier: the larger the company, the more so. The larger the company, the more noise every team has to cut through. These connections help cut through the noise.

Oops, sorry folks! I got too excited about the event and shared too early. Turns out we have a sign up limit and I wasn't supposed to share with the whole internet 😅- I'll repost if we can open it up more broadly.

One of the things that I think is sad about the decimation of Twitter eng is that Twitter was doing a lot of interesting (and high ROI) engineering work that, at younger companies, is mostly outsourced to "the cloud" or open source projects A few examples off the top of my head:

engineers and UI/UX people laid off by twitter, meta and other big tech companies should join life sciences software building efforts. their skills are precious here and can contribute to something more meaningful to humanity compared to adtech and social media.
