New paper alert! ๐nature.com/articles/s44172-0โฆ
Researchers present a cost-effective way to convert a standard commercial microscope, present in many imaging facilities, into a super-resolution microscopy capable of nanoscale imaging via #DNAPAINT #microscopy.
Congrats to all authors!!

A new DNA-PAINT protocol just dropped!

Published in STAR Protocols, researchers created a step-by-step guide for fixing and stainingย Drosophila melanogasterย embryos for DNA-PAINT imaging of E-cadherin nanoclusters and F-actin organization at cell-cell junctions at the nanoscale.
We're proud that the team recommends our #DNAPAINT ready anti-GFP single-domain antibody (sdAb) as part of their workflow.
Congratulations to all researchers!
Not all DNA is created equal and in super-resolution imaging, handedness matters.
Left-handed DNA (Z-DNA) reduces imaging background because it's structurally distinct from endogenous B-DNA, which is especially useful for nuclear imaging. Regarding multi-target imaging, swapping your existing docking strand for a left-handed version is an easy way to increase target multiplexing capabilities.
Right-handed DNA (B-DNA) is the classic. It's suitable for most targets and the backbone of our #DNAPAINT kits.
We now offer both as custom DNA conjugates with your protein of choice.

๐ฌNew publication spotlight!
๐:pnas.org/doi/10.1073/pnas.26โฆ
While antigen presentation is central to cytotoxic T-cell immunity, how antigens are presented at the nanoscale remains poorly understood.
Jacobs, Menche et al. tackled this question in human dendritic cells using quantitative DNA-PAINT microscopy. Their key finding: peptideโMHC I complexes are overwhelmingly monomeric, with antigen presentation regulated by surface density and spatial compartmentalization rather than higher-order clustering. This reframes how we think about T cell triggering and has consequences for the design of cancer vaccines and immunotherapies.
Our custom DNA-PAINT anti-ALFA single-domain antibody was used to label peptideโMHC I.

The diffraction limit stops here.
250 nm resolution blurs away the biology you're trying to study. DNA-PAINT pushes resolution to ~15 nm. The same targets, resolved at nanoscale.

Stojcic et al. used 3D MINFLUX DNA-PAINT to map Na,K-ATPase clustering with sub-10 nm localization precision directly in mammalian cells. They found a nearest-neighbour distance of ~7 nm, providing direct visual confirmation of Na,K-ATPase dimerization and observed higher-order nanoclusters of up to 21 proteins. Great work!

Researchers at the University of North Carolina Chapel Hill wanted to map 12 nuclear targets, including epigenetic marks, transcription machinery, heterochromatin proteins, and DNA at the nanoscale.

Off-the-shelf reagents weren't going to cut it. They needed Exchange-PAINT secondary nanobodies (i.e., sdABs) with orthogonal docking sequences, minimal background in a dense nuclear environment, and high specificity.
We made them a custom set. The result: a comprehensive multiplexed super-resolution map of nuclear organization that revealed how chromatin compartmentalization emerges at the nanoscale.

Most microscopy techniques eventually run into the same wall: your fluorescent dyes bleach out before you can collect enough data.
DNA-PAINT sidesteps this entirely.
Instead of relying on a fixed dye that degrades, DNA-PAINT uses short, dye-labeled DNA strands (imager strands) that transiently bind complementary strands. Because imagers are freely diffusing in solution and continuously replenished, bleaching is a non-issue.
The result: stable blinking for hours, sub-20 nm resolution, quantitative capabilities, and unlimited multi-target imaging possibilities.
At Massive Photonics we aim to make DNA-PAINT accessible with imaging kits designed for real labs, not just specialist setups.
