Degraders were on the front page of the Friday edition of the WSJ Business and here's a little context for people like my dad who are not bio focused.
Drugs that target proteins usually work by binding the protein of interest and preventing it from working properly. But, degraders work by connecting the protein of interest to a junk processor called E3UL. The degrader brings the junk processor to the disease-causing protein and essentially makes it way more likely that the disease-causing protein is going to get tagged by the junk processor for destruction.
And actually, thalidomide was the first degrader, except we didn't know it at the time. It was offered as a morning sickness antidote in the late 1950s but was later recalled after it caused birth defects. Decades later in the 1990s, it came to be used in multiple myeloma after clinical trials showed it worked well. It was a backbone of treatment. Its derivatives are still the cornerstone of myeloma treatment regimens. Despite its use in the clinic, we didn't really know how it worked and that it was a degrader until 2010. Knowing how a drug works biochemically doesn't always come before figuring out if a drug works for patients.
Back to degraders. Degraders attack proteins that live INSIDE cells. Proteins that cause disease that exist inside cells are infinitely tougher to target than proteins that live on the OUTSIDE of cells. For example, antibodies can attack proteins that live on the outside of cells and we as humans have gotten quite good at making antibodies.
The cell is a place packed full of billions of molecules buzzing around participating in billions of reactions. Degraders bring two things into proximity to force a specific single process to occur amidst the chaos.
Degraders are a specific instance of a general class of drugs called molecular glues that act to bring a disease-causing protein into close proximity with something else in order to treat disease. Daraxonrasib, the pancreatic cancer drug that has doubled overall survival, is a molecular glue that brings together KRASG12X (disease-causing protein) and CYPA so that CYPA blocks access to KRASG12X.

Today I smiled for real for the first time in more than 5 years. Thank you to the jury, judge, court officer and clerk. And of course to my rockstar legal team. I truly wish the best for everyone involved.
A civil jury ruled in favor of ex-MIT scientist David Sabatini, who was accused by a younger colleague of inappropriate sexual conduct in his Cambridge research lab, and awarded him $6 million in damages. trib.al/s3v9VIX

In a randomized trial, starting them within 24 hours of symptoms shortened colds by 44% (7.1 to 4 days) and cough duration by 58%.

Don’t play the victim.
It’s the worst human emotion you can have if you want to be successful.
You have to have a belief in your heart that the universe is bending toward you for good.
That you’re going to attract people who want to help you.
And when something bad happens realize that it’s a speed bump on the road to your destination.
Here’s the mistake most people make: they think life is supposed to go up at a 45-degree angle, stress-free, the whole way.
And then they go on the internet and see filtered people who look like that’s exactly what’s happening to them.
I guarantee you, everybody is living an unfiltered life, you just don’t think so.

Three facts about life I want you to learn now.
1. Nobody cares about you and what other people think of you is none of your business.
People aren’t thinking about you anyway so f*ck em.
2. No one is coming to save you.
Not your parents or your friends. Parents might want what’s best for you, but they worked in a different economy and a different era, so they can’t fully understand the dilemmas you’re facing right now.
3. It’s not one big thing that makes you special.
Most people work their 9 to 5, but they’re thinking about getting home to watch a game or tv show.
Take the next hour and go through your contacts, call 20 people, rebuild a relationship or start a new one.
You’re now ahead of the average person.
In life and business you can’t be 50% in, you can’t even be 90% in. You have to be all in.
Whatever it is, your business, your health, your craft, go the extra mile.
Partial commitment isn’t differentiating enough.
Nobody is coming. Go get it yourself.

This new magnesium study is insane.
Healthy adults taking 2 grams of magnesium daily showed a 7.5-year reduction in cognitive brain age. The score combined working memory, episodic memory, and reaction time.
Here’s the break down (1/10):
Readers added context they thought people might want to know
The study tested magnesium L-threonate (Magtein) specifically—not generic magnesium—in adults with self-reported poor sleep; it was funded by Threotech Inc. (Magtein maker). The promoted product is magnesium glycinate. frontiersin.org/journals/nutri… pubmed.ncbi.nlm.nih.gov/41601871/ amazon.com/dp/B0GS3WYGNS

No question that @IsomorphicLabs and other AI drug discovery initiatives can radically accelerate the process, which today is ridiculously protracted. But the objective or expectation of finding "cures" for all diseases is unrealistic IMHO, no matter how much capital is invested.
