One more CRISPR story we just posted on Biorxiv. Adding 5' modifications to donors stimulates HDR and donor potency. Probably does so by preventing end joining reactions that reduce available donor in cells. Please pass along. biorxiv.org/cgi/content/shor…
Don't forget to optimize your RNP concentration using a co-injection marker! It is toxic, you will still get short edits with ssODNs but you won't get long edits if the conc. is too high. Worms getting long dsDNA seem to be more sensitive to RNP dose.
