Next-generation KRAS G12C inhibitor olomorasib shows promising pan-tumor activity in a first-in-human study. ORR ~37% in non-CRC tumors with durable response. Activity even observed after prior KRAS G12C inhibitors. KRAS targeting keeps evolving. nature.com/articles/s41467-0…

SCLC is not a kinase driven disease. At our 2025 Year in Review @charlesrudin explained why we cannot simply copy the adenocarcinoma playbook. While we spent 20 years targeting driver oncogenes in NSCLC the future of small cell lies on the cell surface. From tarlatamab to ADCs the new wave of efficacy comes from targeting unique surface antigens. Watch the full discussion with @peters_solange and @MartinReck2 ➡️ lungsummit.org/adcs/

Perspective @NatureCancer on GLP-1 medicines and cancer risk. Reducing obesity can reduce cancer risk, but beyond that, GLP-1 receptors are expressed on certain cancer cells - could be good or bad. Real world studies suggest lower incidence of some cancers. Overall, impact on inflammation, TME, and cancer risk needs closer study. nature.com/articles/s43018-0…

In non-metastatic TN-IBC, neoadjuvant chemo + pembrolizumab achieved a 31.7% pCR—lower than expected despite improvement vs historical chemo alone (18%). 94% had surgery; 6.4% progressed during neoadjuvant tx. Open esmoopen.com/article/S2059-7…

Interesting move in the new NCCN guidelines: ADCs are now listed as a first-line option in mTNBC, even before FDA approval. Highlights how rapidly the treatment landscape is evolving. #sabcs25 @ASCO @SABCSSanAntonio #MedTwitter @OncBrothers @TotalHealthConf @PTarantinoMD @drsarahsam

Do HER2 mutant #NSCLC tumors progressing on T DXd lose HER2 dependence? No. Resistance is mainly driven by payload related mechanisms such as SLFN11 loss or ABCC1 gain, or impaired antibody binding due to domain IV mutations, rather than loss of the HER2 driver. HER2 signaling persists, and tumors retain sensitivity to #HER2 TKIs (zongertinib/poziotinib). Progression on #TDXd opens a rational path to TKIs jto.org/article/S1556-0864%2… @OncoAlert @AndresFCardonaZ

Primary vs Metastasis: Genomic Differences In the analysis of 5,692 matched primary–metastatic samples from 2,846 patients in the AACR Project GENIE database: 🔹️Metastatic tumors show a modest increase in mutation burden 🔹️FGA is higher → reflecting metastatic genomic instability 🔹️Eleven genes are more frequent in metastases (MYC, CDKN2A, ESR1, AR, etc.) 🔹️The most pronounced differences appear in lung, breast, colorectal, pancreatic, and prostate cancers. 💬The study shows that metastatic tumors exhibit a limited but biologically interpretable degree of genomic divergence compared with primary tumors (likely under therapeutic selection), with the major separation arising from copy-number alterations and structural variants. The clinical utility of re-biopsy therefore varies by tumor type and by the expected dynamics of metastatic evolution. tandfonline.com/doi/full/10.…
