really excited to share my postdoc work, where we used humanized mice to discover the mechanisms of pancytopenia in patients with DPP9 deficiency, which wasn't recapitulated in conventional mouse models. We reveal a DPP9-CARD8 pyroptosis axis governing survival of human HSPCs.
Nominating CARD8 and Caspase-1 as therapeutic targets for patients with Hatipoglu syndrome: doi.org/10.1172/JCI207530
Human DPP9 deficiency causes severe pancytopenia that is not recapitulated by conventional Dpp9-/- mice. Here, Tianli Xiao @tianli_xiao et al. @YaleMed have develop a new way to model this human genetic disease using CRISPR editing of human HPSCs in mice. The new humanized mouse model recapitulates cytopenia in DPP9 deficiency and reveals why patients with DPP9 deficiency have loss of blood cells.

Pancreatic cancer is notoriously hard to treat, but we are making progress. Recent @theNCI -funded work that sheds light on mechanisms behind #daraxonrasib resistance and could help clinicians choose treatment combos that outsmart pancreatic tumors. go.nih.gov/AcvCu95
So much hope.
