RNA science has changed what medicine can do for patients. It’s as if we've learned a new language for writing medicines - and we are still early in that story.
@SailBiomeds is writing the next chapter. Their In Vivo CAR-T program uses eRNA™ and targeted nanoparticle delivery to instruct the immune system from within, with the goal of achieving durable remission for the hundreds of millions of patients with immune-mediated diseases.
Not managing these diseases, but ending them.
Today's announcement of a strategic partnership with @JNJInnovation is a meaningful step toward bringing this curative potential to patients in need.
Congratulations to the Sail and Flagship teams.
.#FlagshipFounded @SailBiomeds announced today a collaboration and option agreement with @JNJInnovation to advance it’s In Vivo CAR-T platform for immune-mediated diseases.
This milestone reflects years of pioneering work across the Flagship ecosystem—from foundational innovations in eRNA and targeted nanoparticle delivery to the creation of a platform with the potential to unlock an entirely new generation of programmable medicines.

The most meaningful scientific progress starts with a clear understanding of the people it is meant to help. That perspective came through powerfully when Dr. Wendy Chung joined Sail CEO John Mendlein for a chat with the team.
Wendy shared reflections on her path into medicine, the career steps that shaped her work as a physician-scientist, and the importance of keeping patients at the center of scientific progress.
Her reflections were a powerful reminder of why Sail is working to advance new eRNA™ medicines, including our In Vivo CAR-T program for autoimmune diseases, and what more accessible, effective therapeutic approaches could mean for patients and families.

In Vivo CAR-T is one of the most closely watched areas in biotech right now, and for good reason. But, the key challenge is not merely inducing CAR expression in T cells.
The real bar is achieving the pharmacology needed for curative immune reset: reaching the appropriate T-cell populations, in the correct anatomical compartments, for a sufficient duration to drive deep enough B-cell depletion across these tissues to disrupt autoreactive immune circuits to enable cures for autoimmune patients.
@DrugDiscovNews explores how Sail’s approach combines Endless RNA™ with targeted nanoparticle delivery to enable durable but transient pan-T-cell (CD4 and CD8) CAR-T generation in vivo, with the dose efficiency and depth of B cell depletion in tissues needed to support immune reset.

Sail Biomedicines is pleased to welcome Dr. Gene Marcantonio as Chief Medical Officer. Gene joins Sail as we advance our lead in vivo eRNA™-TNP CAR-T program for autoimmune diseases toward the clinic and continue building the clinical strategy needed to translate our science into a potential new approach to treating autoimmune diseases.
His experience across translational medicine, early clinical development, immunology, and therapeutic strategy will help guide this next phase as Sail moves from preclinical science toward clinical execution.

In Vivo CAR-T for autoimmune diseases will be defined by more than expression. At ASGCT 2026 and TIDES USA 2026, Sail will present data from its lead In Vivo eRNA™-TNP CAR-T program highlighting the dose efficiency, tissue activity, and depth of B-cell depletion we believe will define meaningful activity and a potentially curative treatment approach in autoimmune disease.
The presentations will include nonhuman primate and other preclinical findings supporting advancement toward the clinic. They will also demonstrate how Sail’s integrated approach, pairing sustained eRNA-based CAR expression with targeted nanoparticle delivery to both CD4 and CD8 T cells, achieves deep B-cell depletion across blood and tissues, including lymph nodes and bone marrow.

In this Pharmaceutical Executive interview, Vincent Hennemand, Sail’s Chief Product & Operations Officer, discusses why In Vivo CAR-T is drawing so much interest.
Autologous CAR-T has shown what deep B-cell depletion can achieve in autoimmune disease, but broader use will depend on approaches that are simpler, more scalable, and better suited to outpatient care.
Sail combines Endless RNA™ constructs and targeted nanoparticle delivery to transiently program T cells in vivo. The goal is to bring the depth of effect associated with auto-CAR-T into a format that could be more practical and accessible for people living with autoimmune disease.

A little friendly competition to start the day at Sail! With nearly 100 hidden eggs and 4 special prizes, the office egg hunt brought out everyone’s competitive side.
We’re excited to welcome John Lepore, MD, to the Sail Biomedicines Board of Directors. John is a physician-scientist and biopharma leader who has spent his career helping translate breakthrough science into medicines.
As former Head of Research at GSK and now CEO-Partner at Flagship Pioneering and CEO and President of ProFound Therapeutics, he has led global R&D strategy, portfolio prioritization, and platform-driven drug development, experience that aligns closely with Sail’s next phase as it advances its in vivo CAR-T program for autoimmune diseases into the clinic.
At Sail, we are building a new generation of genetic medicines with our Endless RNA™ (eRNA™) product forms and targeted delivery platform. John’s perspective at the intersection of platform innovation, translational science, drug development, and CEO experience will be invaluable.

This week, Sail Biomedicines will present new preclinical data at the Immune Resetting: B-Cell Mediated & Beyond Summit, highlighting the dose efficiency profile of its lead In Vivo CAR-T program.
Michael Mingueneau, SVP & Head of Immunology, will discuss how Sail’s targeted nanoparticles and Endless RNA™ (eRNA™) constructs enable transient in vivo programming of T cells, supporting deep depletion of B cells in blood and lymphoid tissues at low dose levels, including evidence of repopulation toward an immature B-cell state consistent with immune reset.

Circular RNA can last longer, but durability alone is not enough. To become a medicine, RNA must translate efficiently and reach the right cells.
Sail Biomedicines’ Endless RNA™ (eRNA™) platform is engineered for durable, tunable protein expression and paired with targeted nanoparticles designed to deliver RNA to defined cell types. Together, these technologies enable programmable control over both expression and delivery.
In this @DrugTargetRev feature, Ewen Cameron, Head of Platform at Sail Biomedicines, discusses how eRNA constructs and targeted delivery are helping move RNA therapeutics beyond burst-and-fade biology and opening new possibilities in immunology from vaccines to in vivo CAR T cell programming for autoimmune disease.
