mTOR and Chromatin Regulation: An Environmental-Epigenetic Axis In our new preprint, we demonstrate that mTOR, the central regulator of cell growth and metabolism, functions as a chromatin regulator in mouse ESCs.

We find that nuclear mTOR selectively binds to developmental gene promoters in synergy with PRC1-deposited H2AK119ub1. At these sites, mTOR associates with the RNA Polymerase II machinery and coincides with high levels of RNA Pol II pausing.

This interaction may help to maintain master developmental regulators in a "poised" state, suggesting a role for mTOR in linking environmental cues to the timing of gene activation. Read the preprint here: biorxiv.org/content/10.64898…

Can metabolism regulate more than bioenergetics during human development? Our new study says YES! We show that metabolism influences cell fate choice in human naïve pluripotent stem cells, histone acetylation, and trophectoderm maturation. rb.gy/spqr9j @NatureCellBio

The @dhnisz lab with the @VingronLab found chemical features in transcription factors (TFs) that encode submaximal transcriptional activity. The findings in @NatureCellBio hint ways to create optimized TFs that could be used for regenerative therapies. --> molgen.mpg.de/4726976

6/8 A combination of histone mod. CUT&Tag and functional readouts using SSEA-1 revealed a TET-dep. bookmarking of active enhancers and L1Mds that is unique within the chromatin landscape. Indicating a preferential react. upon reentering the proliferative phase after dormancy.

7/8 Using zygotic knockouts, ex-vivo and in-vivo models of diapause we supported the finding that in-vivo dormancy/diapause depends on TET activity. Augmentation of TETs via its known cofactors aKG and VitC increased early survival of blastocysts ex-vivo.

8/8 In summary, this work provides a molecular basis of how pluripotency is protected through a state of dormancy. The bookmarking hypothesis we put forward, provides an intriguing explanation of how cells remain dormant but yet maintain their identity once reactivated.

5/8 Using TF motif enrichment and TF footprinting we unraveled a TF network that potentially safeguards the pluripotent state. However, not via an immediate effect on transcription. But potentially via bookmarking the dormant genome. I.e. TFE3 accumulation at dormancy targets.

4/8 Although TETs are promiscuous DNA-binding proteins, only a fraction of TET peaks is actively protected (demethylated) in dormancy when methylation increases. These protected regions consist of ESC enhancers and L1Md repeats (LINE1).
