The first evidence of in vivo SARS-CoV-2 escape from antibodies: emergent Spike deletion H69/V70 and D796H mutation in a convalescent plasma (CP) treated patient. These mutations conferred reduced susceptibility to the CP and sera from multiple donors. medrxiv.org/content/10.1101/…

Long thread on bioRxiv paper co-authored with #ListerFellow @GuptaR_lab. New mutation patterns in SARS-CoV-2 from patients with chronic infection. Evidence of CD8 T cell-escape and adaptation to tissue compartments outside the upper-respiratory tract. biorxiv.org/content/10.64898…
ORF6 is a SARS-2 protein that shuts down traffic between the nucleus & cytoplasm in host cells, inhibiting host mRNA export & immune responses. But in all variants since early 2023, ORF6's abilities are hampered by the ORF6:D61L mutation. Two newish ORF6 observations.... 1/15

#2: Mutations overrepresented in chronic seqs (relative to circulating seqs) are almost certainly functionally meaningful—e.g. Ab evasion, T-cell evasion, or adaptation to a tissue type. See new preprint by me, Darren Martin, & @GuptaR_lab on this. biorxiv.org/content/10.64898… 7/

Once again, for those not already aware, clinical academics contribute to both the health and wealth of the UK. We need to be supporting them and growing their number not hastening their extinction. #ClinicalAcademicCrisis
There is a crisis in clinical academia in the UK. We talk of the need to expand medical schools, embed research into the NHS to improve care, & the importance of a vibrant life sciences sector for the economy of the UK, & yet we seem unable to tackle the underlying issues 1/9

Finally finished a long-gestating paper on mutation patterns in chronic infections (with co-authors Darren Martin & @GuptaR_lab) & it's now available in preprint form. Paper too long for 1 summary 🧵, but I'll do a cursory overview now, then 🧵s on subsections later. 1/16

Wow just heard about new clinical trials of a new drug for hep B infection at the European HIV hepatitis meeting - antisense oligo nucleotides showing promising results in those already taking nucleotide/side inhibitors. And for hep D a new entry inhibitor.@hkjcghi @MedCambridge

Very concerned about the state of final preparedness in light of New Ebola outbreak and hantavirus cruise ship incident. We are committed @hkjcghi and @MedCambridge to addressing preparedness against a range of pathogens, but funding needs to step up in a consistent way.
