In our newly published study, vitrification media temperature was associated with markedly different blastocyst development outcomes despite comparable survival and fertilisation rates.

🌟 Conclusion
The oocyte cortex is a biomechanical decision-making layer controlling fertility, genome stability, and embryo survival and a powerful future tool for non-invasive IVF diagnostics. #ESHREjc
💉 ICSI = A Cortical Stress Test
Injection funnel shape & persistence reflect cortical health and predict fragmentation, blastocyst quality, and pregnancy outcomes. #ESHREjc
📊 IVF Insight
Mechanical tests show:
Cortical stiffness predicts blastulation better than morphology alone.
Biomechanics > embryologist grading
#ESHREjc
🎯 Cortex Stiffness Tunes Chromosome Fidelity
Too stiff?
Too soft?
→ Both cause spindle failure and aneuploidy.
There is a narrow “Goldilocks zone.” #ESHREjc
⚡ Fyn Kinase = Cortical Master Regulator
Loss of Fyn → defective actin cortex, failed polar body extrusion, abnormal zona pellucida, chromosome errors → subfertility. #ESHREjc
🧱 Actomyosin Engineers the Cortex
ERM proteins+actin+myosin sculpt polarity, actin caps, and spindle migration.
Disruption = misalignment, meiotic arrest, aneuploidy.
#ESHREjc
⚖️ Mechanical Polarity
• Low-tension microvillar domain → sperm fusion
• High-tension amicrovillar domain → spindle anchoring
A 2.5× tension difference controls asymmetric division. #ESHREjc
🌀 Meiosis Triggers Cortical Remodelling
TZPs retract → cortex reorganises → asymmetric forces reposition the spindle → polar body extrusion becomes possible.
Bad cortex = poor embryos. #EHSREjc
🔁 Bi-directional Signalling
Through the cortex:
• CC → oocyte: metabolites, meiotic arrest
• Oocyte → CC: BMP15, GDF9
→ Oocyte competence depends on cortical signalling. #ESHREjc
🦄 Oocytes Are Different
Typical cell cortex: 50–400nm
Oocyte cortex: ++ Thick
➡️ Thickness: Specialised functions.
🤝 Cortex: Communication Hub
Hosts transzonal projections (TZPs): actin bridges that connect cumulus cells&oocyte, delivering nutrients and signals. #ESHREjc
🧶 The oocyte cortex is not just a membrane lining. It’s a specialised micromechanical command centre that controls meiosis, fertilisation, embryo integrity, and IVF success. #ESHREjc
🚨 Is the oocyte cortex the hidden key to egg quality, fertilisation, and embryo development?

I believe that, from a design perspective, it should not be difficult to conduct a retrospective clinical study examining the effect of paracetamol exposure during ovarian stimulation or embryo transfer, in order to obtain clinical evidence.
