Thousands of cocultures in one standard 6-well plate. No microfluidics, no new labware. Watch what your T cells actually do over days, cytotoxicity, persistence, exhaustion, then pull the winners out alive, transcriptome intact. Function first. Markers optional. enrichbiosystems.com

Honored to present our Tarlatamab data in active brain mets & treatment beyond isolated CNS progression at #SNOASCO2026! Key findings in 91 pts: ✅ 33% intracranial ORR in ACTIVE, radiation-naïve brain mets ✅ No clear ↑ in severe CRS/ICANS w/ CNS radiation concurrent with tarlatamab ✅ Treatment beyond isolated CNS progression feasible: +4.3 mo median additional time on therapy with local therapy First report of continuing a BiTE beyond CNS progression in SCLC. Manuscript in process. Strong signal. Prospective validation needed. 🧠🫁 Grateful to the SNO/@ASCO chairs & organizers, Dr Hattangadi-Gluth @JHGLab and Dr Naidoo @DrJNaidoo @DukeCancer @UNC_Health_Care @DukeHealth @DukeMets

CAR-T cell efficacy isn't a single metric, it's a multifaceted puzzle of killing potential, exhaustion profiles, and proliferation. Because the low-hanging fruit of killing has been picked, the next era of immune engineering requires deep screening capable of isolating the subtle, combinatorial variations that truly optimize construct performance. #CAR_T #CellEngineering #celltherapy #cancerimmunotherapy

New data identifies NDRG3 as a previously undescribed regulator of restimulation-induced cell death in T cells, suggesting NDRG3 as a target for autoimmune disorders and chimeric antigen receptor T cell treatment for cancer. Learn more: ow.ly/ftZf50ZhTii.

ADCs as "tumor ecosystem-targeting therapies". The next wave is already here. Excellent review on where the field is heading: target expression alone is insufficient to predict activity and multidimensional biomarkers are needed cell.com/cancer-cell/fulltex… @OncoAlert @BarbaraPistill2

💻 On-Demand Webinar: Linking CRISPR Screen Hits to Functional Phenotypes: Decoding CAR-T Cell Fitness Not all CRISPR hits are equal, some edits drive proliferation, others enhance cytotoxicity or persistence, and bulk screening readouts can't always tell you which is which. In this on-demand webinar, Dr. Brandon Simone (Saar Gill Lab, University of Pennsylvania) pairs a CRISPR/Cas9 epigenetic modifier screen with the TROVO system to distinguish edits driving improved CAR-T19 cell fitness and effector function. By continuously monitoring individual microwells and selectively recovering cells of interest, Dr. Simone moved beyond bulk screening hits to directly observe subtle differences behind improved performance, tracking tumor killing and T cell expansion over time to separate edits that promote proliferation from those that enhance cytotoxicity, persistence, or both. Highlights: -How single-well resolution distinguishes proliferation-driven hits from cytotoxicity- and persistence-driven hits -How functional heterogeneity among top screening hits was uncovered using continuous monitoring -How targeted recovery of high-performing clones connects pooled CRISPR screening to mechanistic follow-up Watch on demand: us06web.zoom.us/webinar/regi… Learn more about TROVO: enrichbiosystems.com #CART #CRISPR #CellTherapy #CancerImmunotherapy #FunctionalGenomics #EnrichBiosystems #TROVO

A pooled CRISPR screen flags genes that boost CAR-T fitness — but a top hit could be improving cytotoxicity, persistence, proliferation, or some mix of the three. Bulk readouts can't tell you which. Dr. Brandon Simone (Saar Gill Lab, UPenn) combined an epigenetic modifier CRISPR screen with single-well functional tracking on TROVO to pull that apart — distinguishing edits that drive expansion from those that actually improve killing or persistence, then recovering the high-performing clones for follow-up. A typical CAR-T infusion contains somewhere between 100 million and 1 billion cells. Scientists spend weeks engineering and growing them. Then they inject them and mostly hope for the best.

Most CAR-T cells infused into a patient never reach the tumor. They get exhausted, lost, or shut down by the immune microenvironment before they can do anything. The ones that do make it are doing almost all the work.

Did you know CAR-T therapy takes 3-6 weeks to manufacture after a patient's cells are collected? For someone with aggressive leukemia, that's a long time to wait. It's one of the reasons the field is so obsessed with making this faster and more reliable.
