We investigated the role of ArgR, the master regulator of arginine metabolism, in Crohn’s disease-associated bacteria. ArgR influences cell invasion, biofilm formation and the expression of multiple virulence-associated genes.

We show that the secondary messenger ppGpp is a key regulator of virulence in AIEC strain LF82.
Using infection models and transcriptomic study, we found that ppGpp controls the expression of multiple virulence-associated factors. In absence of ppGpp, LF82 becomes dramatically less virulent.
Our results position ppGpp as a potential anti-virulence target.
🔬Finally, we identified two candidate biomarkers with potential to discriminate AIEC from non-AIEC isolates, opening new avenues for diagnostics.
