A Different Way to Think About Cancer Immunotherapy: 🧬Most cancer immunotherapies try to strengthen the immune cells attacking a tumor. CAR-T, for example, genetically modifies conventional T cells so they can better recognize and attack cancer cells. 🎀CRG-150 takes almost the opposite approach. It uses a patient’s own regulatory T cells, or Tregs, which normally suppress immune responses and can be co-opted by tumors to help protect themselves. 🧬Using CRISPR, researchers disrupt NCOA3, the gene that produces SRC-3, in those Tregs. The goal is to reprogram some of the tumor’s “bodyguards” so they help organize an immune attack instead of suppressing one. 🎀The mouse data have been striking. SRC-3-deficient Tregs eliminated established breast tumors, produced strong effects in prostate cancer, and later showed antitumor activity in models of glioblastoma, melanoma, and lung cancer. This is still preclinical. CRG-150 has not yet been tested in patients. 🧬One especially interesting feature: the first human trial does not plan to use lymphodepleting chemotherapy beforehand. The strategy is designed to recruit the patient’s existing immune cells rather than replace them. 🎀The first human Phase 1/2a trial is expected to begin later this year and will include: ▪️Metastatic triple-negative breast cancer ▪️Metastatic hormone receptor-positive/HER2-negative breast cancer ▪️Metastatic prostate cancer 👀This is very early science, and mouse results do not guarantee human success.
