Prostate tumours classified as high Decipher and PTEN inactive have a 45% reduction in hazard of death when Doce is added to ADT. This biomarker should be tested in pts considered for triplet therapy.

There was minimal overlap between the Decipher and PTEN classifiers, which enriched for different KEGG pathways.

In exploratory analyses, PTEN inactivity in prostate tumours from patients with metastatic disease associated with shorter survival on hormone therapy and predicted survival benefit from docetaxel, as compared to those classified as PTEN active.

In pre-specified analysis, high Decipher classifier score was prognostic and predicted survival benefit from docetaxel for metastatic cancers (biomarker-docetaxel interaction p=0.039). Direction of effect maintained in both high- and low-volume disease.

Tumour AR signaling associated with longer survival, whilst increased proliferation associated with shorter survival.

We established a resource of clinical-grade micro-array expression data and IHC markers (Ki-67, PTEN) on prostate tumours from 1523 pts randomised in practice-changing phase 3 trials #STAMPEDE @MRCCTU (testing docetaxel or abiraterone), linking with survival data.
