Peer-reviewed now.
Human T cells did show up, did recognise the virus, and still didn't clear it. By week 8 the CD4 cells had dropped their naive identity and gone cytotoxic - TEMRA - with IFN/JAK-STAT only switching on late, nothing like it acutely. Mice without T cells mounted the stronger innate antiviral response. Neither group cleared the virus.
A failure to resolve.

‼️Remember our hypothesis of autoimmune hypophysitis in ME/CFS and Long COVID, published in 2024?
We proposed that infection-driven immune hyperactivation, genetic susceptibility and loss of immune tolerance could contribute to pituitary dysfunction, ACTH deficiency and hypocortisolism.
Now, the GEM genetic study reports the pituitary gland as the most enriched tissue.
This does not confirm autoimmune hypophysitis, but it strengthens the rationale for investigating a possible neuroimmune-endocrine mechanism in a subgroup of patients.

Replying to @mecfsskeptic
4) One of the more interesting findings is is that the pituitary (green bar below) now stood out clearly in the tissue analysis. This may point to an important role for hormones in the disease.

This could be groundbreaking- a miracle for those fighting chronic illness.
#LongCovidAwareness
An ”immune reset” using modified cells could offer a permanent cure for millions living with severe autoimmune diseases.
A groundbreaking medical shift is underway, as researchers have repurposed cancer-fighting technology to tackle chronic autoimmune diseases, potentially offering a functional cure rather than lifelong symptom management.
By adapting Chimeric Antigen Receptor (CAR) T-cell therapy, scientists can now genetically reprogram a patient's own T-cells to target and eliminate the rogue B-cells responsible for driving autoimmune destruction. This approach acts as a literal 'immune reset'.
In clinical trials, patients suffering from debilitating, overlapping conditions like lupus, systemic sclerosis, and myositis have achieved complete, drug-free remission after just a single infusion, experiencing restored mobility and the total disappearance of autoantibodies.
Traditional treatments for autoimmune conditions rely on heavy, lifelong immunosuppressive drugs that broadly dampen the body's defense systems, leaving patients vulnerable to infections and serious side effects. In contrast, this new era of cell-engineering platforms and highly targeted therapies aims to correct the root cause of the illness.
Emerging trials, including those utilizing mRNA-based CAR T-cell techniques and inverse 'tolerogenic' vaccines, are showing unprecedented success in retraining the immune system to tolerate the body's own tissues.
While long-term data is still being compiled, the initial results represent a paradigm shift, transforming chronic, life-altering conditions into treatable—and potentially curable—maladies.

For MCAS and Long COVID, Japan has a Solution called Ketas (ibudilast) since 1989. The same thing is now in Western trials for neuroinflammatory disease.
Ketas is a brain-penetrant inhibitor of PDE4 (phosphodiesterase 4, the enzyme that breaks down cAMP in immune cells) and PDE10 (phosphodiesterase 10, which breaks down cAMP and cGMP in neurons), with additional activity at PDE3 and PDE11, two more enzymes that clear the same cyclic nucleotides.
It is also an allosteric inhibitor of MIF (macrophage migration inhibitory factor, a cytokine that keeps inflammation from resolving) and an antagonist at TLR4 (toll-like receptor 4, the sensor for endotoxin and damage signals).
Japan approved it for asthma and for dizziness tied to poor cerebral blood flow. The FDA has not approved it. MediciNova runs it as MN-166, with Fast Track and orphan status in ALS (amyotrophic lateral sclerosis), and it has been taken into lingering-COVID protocols for the same glial and cytokine reasons.
MCAS:
Mast cells keep cAMP (cyclic adenosine monophosphate, the signal that tells the cell to stay quiet) low through PDE4.
Low cAMP favors degranulation, TNF (tumor necrosis factor, a core inflammatory cytokine), IL-6 (interleukin-6, the fever and fatigue cytokine), and the leukotriene arm.
Ibudilast raises cAMP by blocking that enzyme, so the cell is less willing to dump. Older pharmacology also gives it leukotriene-receptor antagonism, which is the part an H1 blocker (histamine H1 receptor blocker) never covers.
MIF is released by mast cells and then calls more inflammation in. Blocking MIF shortens that loop. TLR4 is how endotoxin and mold fragments prime the mast cell before a food arrives.
Ibudilast antagonizes TLR4 and cuts the MyD88 (the adaptor that carries the TLR4 signal inward) and NF-kB (nuclear factor kappa B, the transcription switch for inflammatory genes) traffic underneath it.
Long COVID:
The persistent form is not a missing antihistamine. It is microglia that stayed on, TLR4 still ligated by residual inflammatory signals, and mast cells firing on a lowered threshold. Ibudilast crosses into the brain, suppresses glial activation, and raises cerebral blood flow.
That is the cognitive half: processing speed and the wired-but-exhausted feeling track perfusion and glial noise more than they track dopamine.
MIF and TLR4 are the systemic half. They keep the cytokine set point high after the virus is gone. PDE4 blockade in immune cells pulls TNF, IL-1β (interleukin-1 beta, the inflammasome cytokine), and IL-6 down.
People whose long COVID looks like MCAS, POTS-adjacent flares (postural orthostatic tachycardia, the heart-rate jump on standing), brain fog, and post-exertional inflammatory crashes are the overlap this molecule was built for, even though the Japanese label never used those words.

Gen X is currently stuck in the brutal vice grip of the "Sandwich Generation," and it’s completely reshaping their priorities.
You’re 49. Your kids are 17 and about to leave the nest forever. At the exact same time, your parents are 79 and rapidly losing their memory, mobility, and independence.
You sit in a sterile conference room on a Thursday listening to a 32-year-old consultant talk about "cross-functional synergy," and you realize you have maybe five good summers left with your parents and six months before your teenager moves out.
The idea of trading another 50 hours a week to chase a "Senior Vice President" title stops sounding prestigious and starts sounding like an absolute tragedy.
Gen X isn't burning out because they're tired of working. They’re opting out because they finally calculated the real cost of being absent.

Here is my hot take, the US didn’t aid the Saudis hoping the House of Saud falls, US sends massive and devastating munitions to Israel, Isreal flattens Yemen, Saudi and then Iran (greater Israel achieved) US gets all the oil and the Bible thumpers wait for Jesus - Hallelujah

You can always count on us.
It’s called “oil for security”.
Why would I deceive you?
This is the second time the Saudis see that their security relationship with the U.S. provides no real protection—the first time being when Washington did nothing after the attack against the Armco facility in Abqaiq-Khurais in 2019. This will only consolidate a Saudi sense that after the war with Iran new relationships have to be devised to protect the kingdom: nytimes.com/2026/09/14/world…

New Zealand is deep in a brutal Flu A wave right now and the stories coming out are wild.
“Children with influenza A that is evolving into horrific things like severe respiratory failure, encephalitis, guillian barre syndrome… Its not just a flu.”
“They have an unprecedented number of children in there at the moment with flu… some of them do sadly die in there.”
“It has felt worse than Covid and I was hospitalised during lockdown with Covid.”
“One of my classes today had nearly half the kids out due to illness… I don’t remember ever seeing it this bad before.”
“My son’s high school had just under half students away last week sick. It’s a school of thousands!”
Hospitals code red, clinics overwhelmed, people still showing up to work coughing up bright yellow phlegm like they are heroes.
If this is what the southern hemisphere Winter looks like, the northern hemisphere better get ready.
