Our work on #LongCovid 🩸 is now out in @ScienceMagazine!
We found increased complement activation and markers of thromboinflammation in patients with active Long Covid.

Altogether, an overly activated complement system may connect different hypotheses on Long Covid pathomechanisms.
While the results require further validation in larger patient cohorts, they provide a basis for new diagnostic solutions and possibly targeted treatments.

By measuring antibodies against >80’000 viral epitopes we found increased IgG against #herpesviruses in Long Covid patients, particularly against CMV and EBV.
Anti-CMV was associated with markers of antibody-mediated complement activation.

But why is complement activated in the first place?
1) Tissue injury can be caused by, and activate, complement 🔄
2) Cross-talk with a dysregulated coagulation system ↔️
3) Classical activation mediated by antibodies ⬇️

Using #MassSpec, we found increased markers of tissue injury, including red blood cells, platelets, and endothelial cells.
#FlowCytometry paired with #scRNAseq showed increased monocyte-platelet aggregates in Long Covid patients.

Functional assays revealed increased complement activation. Increased C5bC6 serum levels contrasted with the decrease of C7 complexes.
This pattern suggested increased incorporation of terminal complement complexes in cell membranes.

Experimental validation revealed that C7 complexes were decreased in the blood of Long Covid patients. Long Covid patients that had recovered before 6-month follow-up, showed normal levels.

We measured >6’500 proteins in the blood of COVID-19 patients during initial infection and 6 months later, compared to healthy controls.
6 months after infection, one protein stood out: complement C7.

The complement system is part of the innate immune response and is unfortunately often forgotten right after the immunology exam.

Our most important #COVID19 work so far: @SBruningk @JulianeKlatt & @MadStange examined determinants of SARS-CoV-2 transmission in a city, which could guide vaccination strategies. Spearheaded by @AppliedMicrobi2, w/ H. Hirsch & @UniSpitalBasel @ETH_BSSE medrxiv.org/content/10.1101/…

What impacts transmission within cities? Knowledge can guide vaccine strategies -> high populated and mobile areas, but also people with more severity. Read more here: medrxiv.org/cgi/content/shor…
